The cycle starts at the basement membrane, where keratinocyte stem cells divide. One daughter stays to keep the pool stocked; the other begins a one-way journey. Through the spinous layer it enlarges and fills with keratin filaments. In the granular layer it packs for the end of its life: filaggrin granules and lamellar bodies loaded with lipid precursors.
Cornification is the transition. The cell extrudes those lamellar bodies, dismantles its own nucleus and cross-links a tough protein envelope beneath its membrane. What remains is a corneocyte — flat, dead, packed with keratin — surrounded by lipid lamellae formed from what it released. It rides upward until the studs anchoring it are cut, a step covered in desquamation.
