From vitamin to coenzyme
Niacinamide has no receptor of its own. Skin cells take it up and run it through the salvage pathway to nicotinamide mononucleotide and then NAD+, the carrier that dehydrogenases, DNA-repair enzymes and sirtuins depend on. Its phosphorylated relative NADPH supplies reducing power for fatty acid and cholesterol synthesis and for regenerating glutathione.
That upstream position explains the scattered-looking list of effects. Anything a keratinocyte does that costs redox currency — building barrier lipids, repairing DNA after UV, holding its antioxidant pool reduced — draws on the pools niacinamide feeds. It is a supply-side ingredient rather than a switch for one pathway, which is why its effects are broad but rarely dramatic.
What the downstream effects imply
The barrier effect is the best documented. More ceramide, cholesterol and free fatty acid means a better-organised lipid matrix and measurably less water loss, but those lipids must be made and packed into outgoing cells, so the change tracks epidermal renewal and shows over weeks rather than days.
The pigment effect runs on a different mechanism from most brighteners: niacinamide interferes with the handover of melanosomes from melanocyte to keratinocyte rather than with tyrosinase. It acts on pigment about to be delivered, not on melanin already in the upper epidermis, which is why fading is gradual and why it combines sensibly with agents acting earlier. Pigmentation of unclear cause, and persistent acne or rosacea, warrant a clinician's assessment.
Reading the percentages and the pairing myth
Most published work on barrier and tone uses roughly 2 to 5%. Higher strengths are widely sold and generally tolerated, but direct comparisons of 5% with 10% are few, and there is no good evidence that doubling the figure doubles the effect, while reports of stinging rise with concentration. Choosing 5% costs little that has been demonstrated.
The claim that niacinamide and vitamin C cancel each other traces to old chemistry in which the two, in water at elevated temperature, formed a coloured complex and released nicotinic acid, a flushing agent. Those conditions — sustained heat, dilute solution, long contact — are not what a finished serum on skin provides, and modern formulas combine them routinely.

Many studies use concentrations around 2 to 5 percent, and higher amounts do not reliably add benefit and may increase the chance of irritation in sensitive skin.
Can niacinamide be used with vitamin C?
Yes; despite older myths, modern formulations of niacinamide and vitamin C can generally be used together without cancelling each other out.
Why does niacinamide sometimes cause flushing?
True flushing is a niacin effect, mediated by prostaglandin release, and niacinamide does not normally cause it. Warmth or transient redness after a high-strength product usually reflects irritation, or a trace of nicotinic acid present as an impurity.
How long before anything is visible?
Barrier and comfort changes tend to appear within two to four weeks, following epidermal renewal. Tone changes are slower, generally eight to twelve weeks, because the pigment already in place has to be shed.
Is niacinamide the same as niacin, or as an NAD supplement?
Related, but not interchangeable. Niacin (nicotinic acid) causes flushing; niacinamide does not. Oral NAD precursors act on whole-body metabolism and have not been shown to reproduce topical niacinamide's effects on the barrier.
Do the effects last if you stop?
Largely not. The barrier lipids and reduced water loss depend on ongoing supply, so measurements drift back over weeks once use stops. Pigment already cleared does not return unless the original trigger continues.
