Collagen

Collagen — the most abundant protein in the body and the main structural component of the skin's dermis, forming a fibrous network that gives skin its firmness and strength. Produced by cells called fibroblasts, it provides the scaffolding that supports overlying tissue, and its gradual decline contributes to visible signs of aging.

How fibroblasts build it

Collagen starts inside the fibroblast as procollagen: three chains built on a repeating glycine-X-Y pattern rich in proline and lysine. Before they can wind into a stable rope, prolyl and lysyl hydroxylases modify many of those residues. Those enzymes depend on vitamin C, and without it the helix is unstable — the underlying fault in scurvy.

The finished helix is secreted, trimmed at both ends, and assembles outside the cell into fibrils that bundle into fibres, locked by cross-links from lysyl oxidase. The bundles are thick and interwoven in the deep dermis, finer in the papillary layer beneath the epidermis. It is the network, not the molecule, that resists pulling.

What the network does for skin

Collagen is what stops skin tearing when stretched and what holds its shape in between. The same mesh supports the vessels, nerves, glands and follicles running through the dermis, and traps the water-binding ground substance that keeps the tissue plump.

When it thins and fragments the results are visible rather than abstract: skin sits less taut, expression lines set into fixed creases, the surface looks thinner, and small vessels bruise more easily as support falls away. Disorganised architecture matters as much as the total amount.

What wears it down

Intrinsic ageing lowers production gradually. The figure usually quoted is about 1% a year from the mid-twenties — a population average rather than something measurable in one person, and it varies with genetics and body site. Dermal collagen also tends to fall faster in the first years after menopause.

Extrinsic factors act faster. UV raises matrix metalloproteinase levels within hours, so breakdown outruns synthesis, and the resulting fragments themselves signal fibroblasts to make less — a self-reinforcing loop behind much of what is called photoageing. Smoking and chronically high blood sugar push the same way.

Does applying collagen to the skin replace lost collagen?

Topical collagen molecules are generally too large to penetrate into the dermis where collagen resides, so they mainly act as surface hydrators. Ingredients like retinoids and vitamin C are more associated with supporting the skin's own collagen production.

What causes collagen to break down over time?

Collagen declines with natural aging and is broken down faster by UV exposure, which triggers enzymes that degrade it. Lifestyle factors like smoking can also contribute.

Do collagen supplements do anything for skin?

The evidence is mixed rather than settled. Swallowed collagen is digested into amino acids and short peptides, and while some trials of hydrolysed peptides report modest gains in measured elasticity and hydration, others find none. Many positive studies are small, short and industry-funded.

Is collagen the same thing as gelatine?

Gelatine is collagen heated until the triple helix unwinds: the same amino acids, a different structure. Hydrolysed collagen, sold as peptides, is gelatine broken down further. None of the three is absorbed and reassembled into skin collagen directly.

Why is vitamin C linked to collagen?

It is a required cofactor for the enzymes that stabilise the triple helix. Severe deficiency produces scurvy, with bleeding gums and poor wound healing, because the collagen made is defective. It does not follow that intakes above the adequate level produce proportionally more.

Can you measure how much collagen your skin has?

Not in any routine way. Research settings use biopsies, high-frequency ultrasound or optical imaging, none of them everyday tools. How firm skin looks is a poor proxy, since hydration, underlying fat and muscle all contribute to the same appearance.
