Where the difference lies

Retinol is converted, in two steps, to retinoic acid, which binds nuclear receptors and changes which genes skin cells read. The visible consequences — faster turnover at the surface, more procollagen in the dermis, fewer of the enzymes that degrade collagen — are well documented, and so is the cost: a stretch of dryness, flaking and sensitivity while skin adapts.

Peptides do not enter that pathway. A signal peptide is a short amino acid sequence resembling a fragment of broken-down collagen, and skin interprets its presence as evidence of damage and responds by building. The effect is real but generally smaller and slower than a retinoid's, and it varies a great deal between peptides, most of which have thinner evidence behind them.

Which to use, and how to layer them

For visible change in texture, fine lines and uneven tone, retinol is the stronger tool and the better-studied one, provided your skin will accept it. Peptides are the sensible choice for maintenance, for a compromised or reactive barrier, and for anyone who has tried retinol carefully and could not settle. If skin is irritated now, peptides are the safer step while it recovers.

The two are frequently combined, and there is no evidence that peptides blunt retinol. In practice, apply the thinner peptide layer first and retinol over it, or keep peptides for the morning and retinol for the night — the second arrangement also suits a barrier that is still adjusting. Prescription retinoids are a separate matter and a prescriber's decision.
